Construction ProtocolHuman primary islets (85–95% pure) were purchased from Prodo Laboratories Inc (Irvine, CA, USA) and cultured for 4 days in Prodo Islet Media Standard PIM(S) to recover after arrival. Islets were dissociated into single-cell suspension and viable individual cells were distributed by FACS into 384-well plates containing lysis buffer. Spike-in RNA from the External RNA Controls Consortium (ERCC) (Ambion, Life Technologies) was added to each well in 1:40 000 dilution. Single-cell RNA-seq libraries were generated as decribed in Picelli et al., 2014, Full-length RNA-seq from single cells using Smart-seq2, Nature Protocols.
