Abstract |
This study is based on the importance of stem cells in future medicine and the need to understand the fundamental mechanisms that regulate their potential to differentiate towards specific lineages. We aim to gather the information required to understand the regulatory networks of key stem cells and .. [more] more specifically, to understand the transitions between stem cell states using embryonic stem (ES) cells and a recently described intermediate, Epiblast stem (EpiS) cells. We aim to characterize transcriptional changes in mouse embryonic stem (ES) cells and mouse epiblast stem (EpiS) cells following the knockdown of essential genes identified in genome-wide RNAi screens. ES or EpiS cells were treated with esiRNA for 125 candidate genes and total RNA was collected fro .. [more]m two biological replicates 72 hours post-transfection. The RNAs were normalized and arrayed in 96-well plates (500ng in 50 microlitres).
