Abstract |
An oligonucleotide-based enrichment array covering ~1 Mbp of Aegilops sharonensis coding sequence was used to carry out exon capture and sequencing on wheat and three Ae. sharonensis introgression lines in wheat. The latter include CS(4B)4Ssh which is a 4B substitution line in wheat; T4B-4Ssh which .. [more]is a line carrying a homozygous translocation of Chr 4SshL on Chr 4BL in wheat; and T4B-4Ssh#1 which is an EMS-mutagenised version of T4B-4Ssh containing a previously-identified mutation in the 'breaker' element of the gametocidal locus present on 4SshL. After de-novo assembly of the T4B-4Ssh, it was used as the reference genome to map the reads from the other 3 lines and subsequently identify SNPs in the EMS line to identify candidate genes for the 'breaker' element in the gametocidal loci of Ae. sharonensis.
