Roots were exposed for three hours and subsequently 1 cm root pieces were cut just above the root tip and snap-frozen in liquid nitrogen (~15 zones per replicate). For all experiments plants were grown in an environmentally controlled growth chamber at 20°C with a 16h-light/8h-dark cycle and 70% relative humidity.         <LIBRARY_CONSTRUCTION_PROTOCOL>Medicago truncatula (cv. Jemalong A17) wild-type and Mtcre1-1 mutant (Plet et al., 2011: Plant Journal 65:622-633) seedlings were grown vertically for three days on modified Fåhraeus medium (Fåhraeus, 1957: Journal of General Microbiology 16:374-381) agar plates with low nitrate (including 0.2 mM Ca(NO3)2) supplemented with 1 μM amino ethoxyvinylglycine (AVG). Sinorhizobium meliloti 2011 Nod factors (~10-9 M) or water control was pipetted on top of the root susceptible zone. Roots were exposed for three hours and subsequently 1 cm root pieces were cut just above the root tip and snap-frozen in liquid nitrogen (~15 zones per replicate). For all experiments plants were grown in an environmentally controlled growth chamber at 20°C with a 16h-light/8h-dark cycle and 70% relative humidity.
