      <STUDY_ABSTRACT>Live Lin-/lowc-Kit+ singlets from the cranial dura, femur, and skull of adult mice were sorted for whole-transcriptome analysis. Overall design: Leukocytes were mechanically isolated from the cranial dura, femur, and skull of ten 8 ~ 12-week-old C57BL/6 mice after intracardiac perfusion. Cells from the same compartment were mixed together and stained with DAPI and fluorescence-conjugated antibodies against c-Kit and lineage markers. Then DAPI-Lin-/lowc-Kit+ singlets were sorted and subjected to 10x Genomics single-cell RNA sequencing (scRNA-seq).</STUDY_ABSTRACT>
