      <STUDY_ABSTRACT>We engineered human hepatocyte organoids to study the effect of different fibrolamellar carcinoma mutations. To study the effects and differences between the different mutations, we performed bulk RNA-sequencing to understand the transcriptomic changes. Overall design: Multiple clonal lines per genotype (DNAJB1-PRKACAfus, BAP1KO, PRKAR2AKO and double BAP1KO;PRKAR2AKO) were generated and replicates of parental wild type organoids are included, all were cultured in hepatocyte medium. For wild type organoids and double mutant BAP1KO;PRKAR2AKO organoids samples were also analyzed when cultured in ductal medium.</STUDY_ABSTRACT>
