        <VALUE>It is located on a site that had been surface mined for coal in the 1980s and 1990s, reclaimed following the guidelines at that time, and is now part of a preserve managed by the University of Virginias College at Wise. This pool spontaneously formed in a low-lying area and was not a purposefully constructed retention pond. It is bordered along one side by the shrub Eleagnus umbellate, an introduced/invasive species previously used for surface mine reclamation, which provides partial shade. This body of water supports local fauna and amphibian egg clutches are often found there in the early spring.</VALUE>

        <VALUE>Water collected from the pool was used to produce Pond Agar medium to culture micro-algae, (0% bacto-agar, pH 7.4, buffered with 00M Tris). Microeukaryotes were collected by centrifuging two liters of the collected water at 3000xg for 15 minutes, pellets were resuspended in a small amount of sterile distilled water, spread onto Pond Agar, and incubated at room temperature with natural light (windowsill). Plates were screened every few days using a dissecting microscope to identify small isolated colonies. Colonies were transferred to new Pond Agar plates using a flame sterilized inoculation loop and/or a lab spatula. Isolates were re-screened and transferred over the course of several weeks to produce culture plates with a single alga.</VALUE>
