        <LIBRARY_CONSTRUCTION_PROTOCOL>Cells of the limb muscles were isolated mechanically and enzymaticaly using trypsin centrifuged and resuspended with BioAmf. Using a wide-bore pipette tip cells were thoroughly mixed to get single cells, centrifuged and resuspended with 1X PBS with 0.04% BSA. Two RNA single cell libraries were prepared according to 10x manufacture protocol (Chromium Next GEM Single Cell 3' Library &amp; Gel Bead Kit v3.1, PN-1000121)  using 20,000 input cells per sample (10,000 target cells per sample).
