      <DESIGN_DESCRIPTION>20kb library preparation using ultra-low input modifications (&lt;50 ng of organellar-enriched DNA isolated from 14-day-old wheat seedling leaf tissue samples). The methyl-fractionation (MF) method (NEBNext Microbiome DNA Enrichment Kit; Enrichment by methyl-CpG binding domain) was used to enrich for non-methylated total genomic DNA, which results in mostly organellar (non-methylated) DNA with negligible residual nuclear DNA.</DESIGN_DESCRIPTION>
