Normal hematopoietic progenitors were prepared by c-kit positive cell selection using magnetic beads. cKit positive cells were first infected with retrovirus for the combinations of MYC and HOXA9 or MYC and MEIS1. and transplanted into syngeneic mice. Leukemia cells were prepared from the bonemarrow of leukemia-bearing mice and were cultured ex vivo for a couple of passages to remove normal hematopietic cells. RNA samples of these cells were analyzed by mRNA-seq. Total RNA was prepared using the RNeasy kit The libralies were constructed using a SureSelect Strand Specific RNA Library Prep Kit (Agilent) following the manufacturer's instructions.
