ADAR1 has two isoforms, short p110 and long p150. Although both ADAR1 KO mice and ADAR1 KI mice that harbor the editing-inactive E861A point mutation are embryonically lethal, this lethality can be rescued by concurrent deletion of MDA5. To elucidate the effect of a mutation associated with Aicardi-Goutieres syndrome, we generated knock-in mouse model by inserting an AGS-associated point mutation in a catalytic deaminase domain of ADAR1. Then, RNA editing pattern in brain and lung was compared with wild-type mice. Single-cell RNA-seq libraries were obtained following the 10x Genomics recommended protocol, using the reagents included in the Chromium Single Cell 3' Reagent Kit v3.1. All single-cell RNA-sequencing was performed with an Illumina NovaSeq 6000 with the following setup: paired-end, read1 length 28bp, read2 length 91bp.
