Comment[GEAAccession] E-GEAD-444 MAGE-TAB Version 1.1 Investigation Title TGF-beta1 induces mesothelial-mesenchymal transition in human peritoneal mesothelial cells Experiment Description Human primary peritoneal mesothelial cells isolated from tumor-free human omentum were used. Mesothelial cells were cultured on collagen-coated plates in RPMI-1640 media supplemented with 10% FBS and penicillin/streptomycin until confluence and then exposed to TGF-beta1 (10 ng/ml) in low-serum (1% FBS) for 72 hours. Our studies revealed that mesothelial cells, originally functioning as a barrier to the peritoneum, co-organize the tumor microenvironment in the peritoneal dissemination. In peritoneal dissemination, mesothelial cells consist part of the cancer-associated fibroblasts. TGF-beta1 derived from ovarian cancer cells induces mesenchymal changes in mesothelial cells, which enhanced the adhesion of ovarian cancer cells to the peritoneum and their resistance to platinum-based chemotherapy by interacting with each other. Total RNA was extracted from human omental mesothelial cells using QIAGEN RNeasy Mini kit. A total of 2 micro gram of RNA was used for construction. The libraries were sequenced for 300 cycles. Remove reads containing low quality (Qscore<= 5) base which is over 50% of the total base.
