Biotechnol. 22, 1146-1149 (2004) COMMENT The method first splits the mRNA into two aliquots and its cap structure is enzymatically replaced with two types of synthetic oligonucleotides containing MmeI, a type-IIS restriction endonuclease site, and the XhoI restriction enzyme site. Oligo-capping mRNA is then converted into first strand cDNA with a random adapter-primer. The second strand is synthesized using biotin-bound 5'-primer and random adaptor-primer by PCR. The double-stranded cDNA is then cleaved with MmeI, which cleaves 20 bp away from its recognition site. After the 5'-cDNAs are isolated by binding to streptavidin beads, the two pools of tags are ligated to each other.
