During separation, the midgut side of the resection surface was held between the forceps to prevent the release of midgut contents into the PBS. Next, the resection surface of the midgut was dipped into the buffer for chemical analyses, and the contents were collected by gently pressing the midgut with forceps. The collected midgut contents were extracted in 500 µL of methanol. Then the whole sample was sonicated and centrifuged at 10,000 × g for 5 min at room temperature. The supernatant was collected and adjusted by 50% methanol add 0.1% formic acid. Extract sample was analysed using directly infusion (DI) in the quadrupole Time of Flight (Q-TOF) mass spectrometer coupled to an electrospray ionization (ESI) source (TripleTOF 5600+, AB Sciex Ltd., USA). Sample was injected with a flow rate of 1 µL/min for a total run time of 10 minute. MS/MS spectra in the mass range of m/z 50-1,250 were obtained in positive ion mode.
