title Analysis of transcription start sites of in Symbiodinium minutum
description Construction of transcription start site (TSS) library was performed as follows. Cells were cultured at 25 degrees C in a 12 hours light/dark cycle and collected during a light cycle. Total RNA was treated with BAP for 1 h and TAP for 1h. The RNAs were ligated with the RNA oligonucleotide. After DNAse treatment, poly(A)-containing RNA was selected and used for two library constructions (Mfa_zoox_1st and Mfa_zoox_2nd). Sequencing reactions were performed according to illumina instructions for illumina Hiseq 2000. The reads mapping to the draft genome with the predicted 41,925 genes show potential TSS sites. In addition, we find transcripts of 3,329 genes, which is spliced-leader trans-spriced.
