Implantation to the uterus is a specific phenomenon to mammalian development, which triggers a series of important developmental events through interactions between mother and embryos. It is thought that large scale epigenomic changes take place during peri-implantation development, resulting in dramatic changes in gene expression profiles. However, gene expression analysis of the peri-implantation embryos has been hampered by technical difficulties in handling very small embryos of these developmental stages. Here we isolated embryos of embryonic day 3.5 (e3.5) to e6.5 and separated embryonic tissues that constitute the embryos; e3.5 embryos consist of inner cell mass (ICM) and trophectoderm (TE), while e5.5 embryos comprise epiblast, visceral endoderm, and extraembryonic ectoderm. We amplified cDNA obtained from each sample and performed RNA-Seq analysis aiming to elucidate transcriptome dynamics during peri-implantation stage mouse embryos. |
