title Repetitive DNA methylome analysis by single cell shotgun bisulfite sequencing
description We produced shotgun bisulfite sequencing (SBS) data from single murine female primordial germ cells and fully-grown oocytes by an improved multiplexing and amplification-free post bisulfite adapter tagging method (scPBAT). We mapped the low-coverage SBS short-reads of mouse to consensus sequences of repetitive elements. The scPBAT data quantitatively provided over 5x sequencing coverage for at least 30 repeats, and the individual methylation patterns detected were similar to the bulk-cell-based results. Our new single-cell methylome sequencing technique is cost-effective and will help researchers to investigate intergenic methylation changes using limited amounts of mammalian cells as well as cells from other organisms with the genomic annotations.
