Fission yeast cells containing Ch16 minichromosome were randomly mutagenized, and the mutant clones that showed decreased rate of gross chromosomal rearrangements were selected. After crossing against the parental wild-type strain, genome DNA was prepared from around 10 progenies from each candidate, assembled, and subjected for whole genome sequencing. |
