Genome sequencing of Psychotria homalosperma was performed by usingRoche 454 reads. Specifically, total genomic DNA of Psychotriahomalosperma was extracted from fresh leaf collected from Sekimon(26°40'11.3"N 142°09'16.4"E) on Hahajima Island of the Bonin Islands,using the DNasy Plant Mini Kit. The extracted DNA of P. homalosperma waspyrosequenced using a 454 GS Junior System. Multiplex Identifier (MID)tags were used for multiplexing of this sample and another species inthe Bonin Islands, then both samples were mixed. Row data weredemultiplexed and MID tags removed from the reads using Newbler. |
