Grapevine (Vitis vinifera) is an important horticultural fruit trees used for table fruit or wines, etc. This research performed transcriptome analysis (RNA sequencing) to characterize Japanize wine grape cultivar "Koshu" against European wine grape "Pinot Noir", as a reference, whose genome has already analyzed. Total RNA was extracted from the stems and leaves of Koshu and Pino Noir (each 3 samples) using NucleoSpin RNA Plant Kit with Fruit-mate for RNA Purification (Takara), and a library was prepared using the kit. Library preparation was performed using NEBNext Ultra RNA Library Prep Kit for Illumina (New England Biolabs). Sequencing was performed using illumine HiSeq 2500 (Illumina) and bcl2fastq2 conversion software (Illumina, version 2.1) to comprehensively analyze differential gene expression in shoot (leave and stem) between Koshu and Pinot Noir. All samples showed more than 20 million read data. Thus, the sequence data can be used for RNA-seq analyzing. |
