description Sequencing DNA of Buchnera aphidicola, a symbiotic bacterium of a pea aphid, after whole genome amplification (WGA) with phi29 using a single molecule sequencer. Comparing several conditions (two WGA kits, two amounts of template DNA and two DNA treatment methods after WGA process. The results showed that the amplified DNA also can be sequenced by singe molecule sequencer and generate comparable de novo genome assembly result to non-amplified DNA. This method will contribute de novo genome analysis of non-model organisms, from which only small amount of DNA is available.
