title Assembly of Diospyros lotus pooled BAC in ASTRINGENCY locus
description Across persimmon (Diospyros kaki; 2n = 6x = 90) cultivars, pollination constant and non-astringent (PCNA)-type cultivars stop accumulating proanthocyanidins in the early stages of fruit development, with a genetic control by a recessive allele at a single locus, referred to as ASTRINGENCY (AST). In order to construct reference sequence in AST locus, chromosome walking using D. lotus (2n = 2x = 30) BAC library was conducted. It started from a flanking region of a marker co-segregating with AST in D. kaki (Kanzaki et al 2010). The screened 19 BAC clones were pooled into three groups, and sequenced using Pacbio RSII technology. Each BAC pool was separately assembled using HGAP2.0 program. The resultant contigs were ordered using the BAC-end sequences, and further summarized into longer contigs based on the overlap with each other.
