Murine-derived feeder cells - either SNL76/7 feeder cells (SNLs) or mouse primary embryonic fibroblast feeder cells (MEFs) - are widely used for the undifferentiated culture of human induced pluripotent stem cells (hiPSCs). However, it is still unclear whether these different culture conditions impact the efficiency of definitive endoderm (DE) differentiation from hiPSCs. Thus, to investigate molecular pathways in hiPSCs affected by cultured on SNLs or MEFs, we perform the next-generation sequencing analysis of transcriptome in hiPSCs line 201B7 cultured on SNL or MEF feeder cells, and then these data are used for molecular network analysis by KeyMolnet software. The goal of project is to find the proper method of undifferentiated state mentenance of the hiPSCs to induce the efficient DE differentiation. |
