Microbial community structure changes of onsite groundwater treatment system was evaluated. Total DNA were extracted from samples 0.1 of g wet weight (Day 0 and 430) by using FastDNA SPIN Kit for the Soil (MP-Biomedicals). The V4 hyper variable region of the bacterial 16S rRNA gene was amplified by PCR using primers Eub-515f and Eub-806r. The PCR reaction comprised 20 cycles. Then, the amplified products were submitted to next-generation sequencing using an Illumina Miseq gene sequencer at a commercial sequencing service. |
