Rotavirus population shows the dynamic distribution of mutant swarms (termed as quasispecies) due to the accumulation of mutations in the viral genomes caused by lacking the proof-reading function of error in RNA-dependent RNA polymerase. However, there are few evolutionary information about rotaviruses such as evolutionary rates of each genome segment. To accumulate evolutionary information about rotaviruses, applying serial passages of rhesus rotavirus (RRV) after 100-fold dilution in duplicate (1st and 2nd lineages), we then adopted new generation sequencing (NGS) analysis. To calculate the evolutionary rates (substitutions/site/cycle) of 11 genome segment using Markov chain Monte Carlo methods, we needed information about single nucleotide polymorphisms. |
