We have characterized links between the SpdC protein and the WalKR two-component system. We have generated an spdC-deleted strain. This strain behaves like the WT parental strain, at least concerning its growth rate in TSB rich medium at 37°C under aerobiosis. The WT and spdC derivatives were grown in TSB rich medium until mid-exponential phase (OD600nm=1) and RNA were extracted for further RNAseq analysis. Three biological replicates were analyzed for each strain. Using a cut off fold change higher than 2 with a p value less than 0.05, we found that 42 genes were repressed in the spdC mutant strain and 65 were activated. Among them, 24 were previously identified as WalKR-regulated genes, thus confirming our hypothesis concerning the role of SpdC in modulating the WalKR regulating activity. In particular, we have shown that SpdC in involved in regulation of expression of 4 genes encoding cell wall hydrolases and 10 virulence genes. |
