We propose to establish the budding yeast as a model for human GWAS populations. We have previously generated an artificially outbred S. cerevisiae population by crossing four founder strains representative of the major diverged lineages for 12 generations (F12 pool). In addition to bulk analysis of segregants currently ongoing, we are looking to create a very large population of diploid segregants (N ~ 10,000) with known genome sequence and several measured quantitative phenotypes. This will establish a yeast population to model human cohorts used in GWA studies, with an opportunity to follow up mapped QTLs using genetic manipulation. |
