description We combined chemical and enzymatic techniques to map single-stranded DNA (ssDNA) across the mammalian genome by high-throughput sequencing (ssDNA-Seq). The new approach makes use of KMnO4 treatment, which modifies ssDNA in living cells by oxidizing pyrimidine bases with a marked preference for exposed thymidine residues.
