Hinoki sapling was planted in a plastic pot and loosely fixed to a stake using wires. The sapling was artificially inclined after the initiation of cambial growth in May. Sampling was conducted during the most active period of cambial growth in June. After bark removal, differentiating xylem tissue was scraped from the vertical stem of the sapling. For SOLiD sequencing, total RNA was extracted using RNeasy Plant Mini Kit (Qiagen) and treated with RNase-free DNase I (TaKaRa). |
