Fugen site was located at east end of Kairei field (25°19.2271’S, 70°2.4155’E) and we found active black smoker chimneys in this site. The vent fluid temperature was 304°C. We sampled the chimney and divided into the surface and inside layer. The inside layer was constituted by pyrite. We extracted DNA from each layer and subjected to PCR amplification. The amplified 100 bp of 16S rRNA gene fragments were used for 454 pyrosequencing analysis. |
