This sample is total RNA in the siCTRL-depleted and poly I:C stimulated Hela TO cells.HeLa TO cells (Clontech) were grown in Dulbecco's modified Eagle’s medium (DMEM), supplemented with10% fetal bovine serum (FBS) and antibiotics at 37C in a humidified incubator with 5% CO2.According to the manufacturer’s protocol, cells were transfected with poly I:C using Lipofectamine 2000 (Invitrogen). For poly I:C stimulation, cells were seeded into 12-well cell culture plates followed by transfection using the indicated amounts of poly I:C and Lipofectamine 2000. The siCTRL were transfected into cells using Lipofectamine RNAiMAX (Invitrogen) according to the manufacturer’s instructions. Briefly, siCTRL duplexes (final concentration 10 nM) and cells were harvested 48 hr posttransfection. |
