sample comment  The high-throughput sequencing of 16S rRNA genes at the whole community level provides an almost unbiased profiling strategy for measuring characteristic changes in the relative prportions of the microcosms involved in certain functional process in complex systems, such as soil. There are 27 samples for total DNA pyrosequencing and 84 samples for SIP fractionated DNA. The paddy soil cllected from Yangzhou city of Jiangsu province was used for study. Four treatments were performed including 13C-CH4-labeled microcosms (incubated with 13C-CH4), 13C-Urea-labeled microcosms (incubated with 13C-Urea and 13C-CO2), 13C-CH4+Urea-labeled microcosms (incubated with 13C-CH4, 13C-Urea and 13C-CO2) and 12C-CH4+Urea control microcosm (incubated with 12C-CH4, 12C-Urea and 12C-CO2). The 13C-labeled urea can hydrolyze to ammonia and 13C-CO2, which is benificial for the 13C-label of autotrophic ammonia oxidizer. Pairwise comparison among the treatments of 13C-CH4, 13C-CH4+Urea, and 13C-Urea was used to assess the effect of urea fertilization on methane oxidation, and the role of methane on ammonia oxidation. The soil microcosm with 12C-CH4+Urea amendment was performed as control treatment for the labeled SIP microcosms.
