All animal procedures in the present study were approved by the Committee for Experimental Animals at Zen-noh Embryo Transfer (ET) Center, Hokkaido and the University of Tokyo, Tokyo, Japan. Estrous synchronization, super-ovulation and ET processes were performed as previously described (Ideta et al. 2007). Seven-day embryos (day 0 = day of estrus) were collected from super-ovulated Japanese black cattle. 12 embryos derived from the super-ovulation were transferred non-surgically into the uterine horn of three Holstein heifers (n = 4 each), ipsilateral to the CL on day 7 of the estrous cycle. Elongated conceptuses were collected non-surgically by uterine flushing on day 20. Conceptuses in the uterine flushing media were obtained by centrifugation at 1,000 rpm for 5 min, snap-frozen and transferred to the Laboratory of Animal Breeding at the University of Tokyo. |
