| description |
The worm strain used was KH1752. This is a mutant worm strain. The genotype is unc-75 (yb1701) I. Embryos were obtained by bleaching synchronized gravid adults. The embryos were incubated for 18 hours at 20 degree Celsius in M9 buffer. The synchronized L1 worms were then fed with E. coli strain OP50 suspended in S-complete medium for 3 hours at 20 degree Celsius. The L1 worms were cleaned by washing with the M9 buffer and total homogenate was prepared by grinding the worms in liquid nitrogen. Nuclei were harvested by centrifuging the total homogenate. The nuclei were treated with Nuclear Lysis Buffer and chromatins were harvested by centrifugation. Total RNAs were extracted from the chromatins by using Sepasol I Super (nacalai tesque) and then purified by using RNeasy Plus Mini kit plus DNase I (Qiagen). |
