| description |
The worm strain used was N2. This is a standard wild-type worm strain. Embryos were obtained by bleaching synchronized gravid adults. The embryos were incubated for 18 hours at 20 degree Celsius in M9 buffer. The synchronized L1 worms were then fed with E. coli strain OP50 suspended in S-complete medium for 3 hours at 20 degree Celsius. The L1 worms were cleaned by washing with the M9 buffer and total homogenate was prepared by grinding the worms in liquid nitrogen. Cytoplasmic fraction (clear supernatant) was prepared by centrifuging the total homogenate. Total RNAs were extracted from the cytoplasmic fraction by using Sepasol I Super (nacalai tesque) and then purified by using RNeasy Plus Mini kit plus DNase I (Qiagen). |
