Four-day old Arabidopsis thaliana Col-0 seedlings grown on MS plate (392-00591, Wako) under 12 h white light of 70 to 100 micro mol s-1 m-2 / 12 h dark conditions (LD) were transferred into LL condition. Next day, seedlings were transferred in a 50 mL conical tube at ZT15 (15 h after light on). Five mL of MS liquid containing 2% (w/v) sucrose, and 200 micro M PHA767491 (with a final concentration of 1% (v/v) DMSO), were added into 50 mL tube. Plants were vacuumed for 5 min. Plants were kept for 3 h, and frozen in liquid nitrogen at ZT18. |
